Abstract
We present epi-detected stimulated Raman scattering (SRS) microscopy that employs a water-immersion objective and long-wavelength excitation. The increased numerical aperture and reduced scattering loss enable high resolution imaging with deeper tissue penetration. Furthermore, the inherent spectral response of a silicon photo-diode, which is several orders of magnitude less sensitive to long wavelengths (1200-1300nm), makes filter-less epi-detection possible. The improved SRS system opens up practical biomedical applications such as noninvasive in vivo skin imaging.
