Abstract

Microscopic measurements of oxygen delivery and metabolism are essential for better understanding of cerebrovascular pathologies and interpretation of macroscopic data from imaging modalities such as functional magnetic resonance imaging (fMRI) and diffuse optical tomography. We developed a method to obtain detailed statistics of oxygen content in cortical microvascular networks of anesthetized mice based on two-photon microscopy phosphorescence lifetime imaging of oxygen sensitive dyes. We provide the oxygen intravascular distribution as a function of various morphological parameters such as branching order from pial arteries and veins, as well as the difference in the intravascular oxygen distribution in normocapnic and hypercapnic states.
