Calibration of Ca+ Dissociation Constant of Rhod-2 In Vivo in Perfused Mouse Heart Using Mn+ Quenching

Abstract

We present an analytical model to characterize the calcium dissociation constant of the fluorescent indicator Rhod-2 in vivo in perfused mouse heart using manganese quenching fluorescence transients. We also provide the experimental results to validate this approach and determine that the calcium dissociation constant for Rhod-2 is 720nM.
