Studying Subunit Cooperativity by Counting Hydrolyzed ATP on Single Chaperonin Nanomachines in Solution

Abstract

Single chaperonin proteins are trapped by an Anti-Brownian Electrokinetic trap and hydrolyzed Cy3-ATP counted using stepwise photobleaching. Unlike ensemble averaging the observed ATP number distributions depart from the standard cooperativity models.
