Investigation of 3-D cell-based assays with structured illumination using a digital micromirror device

Abstract

In this paper, we demonstrate a fluorescence optical detection system with structured illumination using a digital micromirror device to investigate various cell-based assays. To study the performance of the optical detection system, we measured 2-D and 3-D assays of fluorescent microbeads and extended the results with mammalian cells. Through image reconstruction after structured illumination, we have improved fluorescence images by 32 times in terms of the size of axial point-spread function. The findings can be useful to supplement structured illumination microscopy that has been actively investigated for various applications.
