Abstract

Raman spectroscopy has been used increasingly to determine the conformation of the peptide backbone and the side chain microenvironment of some amino acid residues. Because of the relative uncertainty about the absolute magnitude of intensity, Raman spectroscopy has been used primarily for qualitative evaluation of properties of proteins. There have, however, been some attempts to use Raman spectroscopy for quantitative purposes in the analysis of protein conformations.
